Artemether injectable injection ,anti-malarial pharmaceutical
Pharmacology Artemether is a derivative of artemisinin, which has a powerful and rapid killing effect on the intra-erythrocytic stage of Plasmodium falciparum, and can rapidly control clinical episodes and symptoms. The mechanism of action of artemisinin is not well understood, mainly interfering with the Plasmodium surface membrane and mitochondrial function. Artemisinin causes changes in the structure of the membrane system of Plasmodium by affecting the ultrastructure of the Plasmodium red inner stage. 2 As a result of the action on the food vesicle membrane, the nutrient uptake of Plasmodium is blocked, and when Plasmodium loses a large amount of cytoplasmic and nutrient material that is not replenished, it dies quickly. The mode of action is through its internal peroxide (hydrogen peroxide) bridge, mediated by free iron produced by hemoglobin decomposition, generating unstable organic radicals and/or other electrophilic intermediates, which then form covalent adducts with Plasmodium proteins and cause death of the parasite. The antimalarial activity of artemether is 6 times greater than that of artemisinin. 3 Toxicology, the LD50 of oral artemether in mice is 895-977mg/kg, and the LD50 of intramuscular injection of artemether oil in rats is 597mg/kg.Sub-acute toxicity test showed that after intramuscular injection of artemether oil in rats at 18mg/kg per day for 14 days, there was no adverse reaction except the decrease in the number of reticulocytes. The drug has some embryotoxicity, manifested by embryonic absorption.
Usage :
1 Adults, usual dose, intramuscular injection, the first dose of 160mg, starting from the second day, once daily, 80mg each time, for 5 days.
2 Pediatric usual dose, intramuscular injection, the first dose of 3.2mg/kg of body weight; the second to fifth days, each time according to the body weight of 1.6mg/kg, once a day.
